Oh well what you can do is make a folder with layer mask around the eye so that you can only draw inside the eye. Then make a layer in said folder, then animate one circle with a black STROKE and a colored FILL from the center of the pupil enlarging it until it is no longer visible in the eye. Then you repeat that 2 or 3 times until you have enough colors and a perfect loop. FINALLY you would make a layer above the folder you made, set it to clipping mask so it can only draw on the eye and add a shine and a bit of a shadow.
