I don't know if DC is still doing this, but to go back to being the insufferable continuity nerd, DC writers had a thing going where Kyrptonite only works on a Kryptonian from the same universe. So Power Girl was even more OP because she's from Earth-2, so the Kryptonite you could potentially kill Superman with has no effect on her.

So to overexplain the joke, she's not effected by the doohickey, she just wants to rev up a Harley.
