I hate to be a french grammar nazi, butit would not be vous etes because she is referring to a single person (or in this case, frog) so it would actually be "Oui, tu es ma matîre". Furthermore, maître is a masculine noun, so it would have to be referred to with a masculine pronoun, so the correct translation would be "Oui, tu es mon matîre".

I'm pretty sure "vous etes" also works as a more formal version of "tu es", and seeing as she's speaking to her new "master", I'm thinking formal language is the way to go ^^
And I think what blows my mind the most is that we have seven images tagged "frog". It's awesome, PC!

She can say "vous êtes" actually. "Vous" can also be used when we're talking to a single person, but an "important" person, someone we have to show respect to.
For example, you can't say "tu" to the president, you have to say "vous".
